PT-141 (bremelanotide) is a melanocortin receptor agonist studied for central sexual-desire pathways. The FDA-approved product, Vyleesi, is indicated only for a specific HSDD diagnosis in premenopausal women; research-grade PT-141 is not that approved product.

PT-141 Research Profile

PT-141, also known as bremelanotide, is a synthetic melanocortin
receptor agonist studied for its effects on central sexual-desire
pathways. FDA approval applies to the specific Vyleesi product and
indication—not to research-grade PT-141.

Product Information

About PT-141

PT-141, or bremelanotide, is a synthetic cyclic heptapeptide
melanocortin receptor agonist. It is related to Melanotan II but is
a distinct compound. Its central melanocortin activity has been
studied in relation to sexual desire and arousal.

Unlike medicines that primarily affect peripheral blood flow,
bremelanotide acts through melanocortin receptors in the central
nervous system. The complete mechanism behind its effect on sexual
desire is not fully understood.

Research-grade PT-141 should not be confused with Vyleesi, the
FDA-approved prescription product. Approval, testing, and the
product instructions for Vyleesi do not apply to research-grade
material.

Research & Approved Use

What Has Been Studied

FDA-Approved Indication

Vyleesi is approved for premenopausal women with acquired,
generalized hypoactive sexual desire disorder (HSDD) that causes
marked distress or interpersonal difficulty and is not due to
another medical or psychiatric condition, relationship problems,
or a medication or drug.

Central Signaling Research

Bremelanotide activates melanocortin receptors, particularly MC4R.
Research has examined central pathways related to sexual desire.
It is not a PDE5 inhibitor and should not be described as simply
improving genital blood flow.

Limits of the Approved Use

Vyleesi is not indicated for men, postmenopausal women, or
enhancement of sexual performance. Use of research-grade PT-141
or use outside the approved indication is not established by
Vyleesi’s approval.

Research Product Distinction

Vyleesi is supplied as a ready-to-use prescription autoinjector.
A research-grade peptide or a compounded or nasal formulation is
not the FDA-approved Vyleesi product.

Evidence Overview

Clinical Evidence and Limitations

Clinical trials supported Vyleesi’s use for its specific approved
HSDD indication in premenopausal women. The results should not be
generalized to people without that diagnosis, to men, or to
research-grade products.

The FDA label states that Vyleesi is not intended to improve sexual
performance. It also does not establish a dose, safety profile, or
effectiveness for unapproved formulations or routes.

Safety & Regulatory Status

Important Safety Information

Nausea is the most commonly reported adverse reaction with Vyleesi;
other reported effects include flushing, headache, and injection-site
reactions. The approved product can temporarily increase blood pressure
and decrease heart rate. Focal hyperpigmentation has also been reported.

Vyleesi is contraindicated in people with uncontrolled hypertension or
known cardiovascular disease. It is not indicated for men,
postmenopausal women, or sexual-performance enhancement.

FDA approval applies only to Vyleesi for its labeled indication.
Research-grade PT-141 is not FDA-approved, and its identity, purity,
sterility, and formulation may differ from the approved product.

For laboratory research only. Not intended for human or animal
consumption, self-administration, diagnosis, treatment, cure, mitigation,
or prevention of disease.
