About FOXO4-DRI
FOXO4-DRI is a synthetic peptide designed using a D-retro-inverso
structure. It has been studied as a senolytic research compound—a
compound investigated for its potential to affect senescent cells.
In preclinical models, FOXO4-DRI was studied for its ability to
interfere with the interaction between FOXO4 and p53. Researchers
proposed that this could allow p53-related apoptosis pathways to
become active in senescent cells.
The D-retro-inverso design was intended to increase resistance to
enzymatic breakdown. This design feature does not establish how the
peptide behaves or how safe it would be in people.

Research Areas
What Preclinical Research Has Examined
Senescent-Cell Pathways
Cell and animal studies have examined whether disrupting the
FOXO4–p53 interaction can affect senescent-cell survival and
apoptosis pathways. Selective effects in preclinical models do not
show that healthy tissues would be spared in people.
Aging-Model Findings
In aged-mouse models, researchers reported changes in measures
including fur density, kidney function markers, physical activity,
and fitness. These are animal findings, not demonstrated human
benefits.
Other Tissue Models
Preclinical research has also explored Leydig-cell and testosterone
markers, lung-fibrosis models, vascular markers, and chemotherapy-
related effects. These findings require further study and do not
establish clinical uses.
Evidence Limitations
No human clinical trials of FOXO4-DRI were identified. Human safety,
effective dosing, long-term outcomes, and efficacy remain unknown.
Cell studies involving human cells are laboratory research, not
clinical trials in people.
Evidence Overview
Preclinical Findings Only
The research so far primarily involves cell studies and animal models.
Results such as changes in kidney markers, activity, tissue fibrosis,
or reproductive markers cannot be assumed to predict outcomes in
people.
Safety, efficacy, appropriate administration, and long-term effects
in humans have not been established. Research results do not support
anti-aging or disease-treatment claims for this product.
Safety & Regulatory Status
Important Safety Information
There is no established human safety database for FOXO4-DRI. Limited
reports from uncontrolled community use cannot determine safety or
predict how an individual may respond. Theoretical effects of removing
senescent cells also remain under investigation.
FOXO4-DRI is not FDA-approved for any indication. Product identity,
purity, and quality may vary between research suppliers.
