5-Amino-1MQ Research Profile
5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule
compound—not a classical peptide—that selectively inhibits
nicotinamide N-methyltransferase (NNMT). Its reported effects come from
cell and animal research; human efficacy and safety have not been
established.

Product Information
About 5-Amino-1MQ
NNMT is an enzyme that uses S-adenosylmethionine (SAM) to methylate
nicotinamide, a precursor involved in NAD+ metabolism. NNMT is
expressed in adipose tissue and has been studied in connection with
obesity-related metabolic changes.
By inhibiting NNMT, 5-Amino-1MQ has been investigated for effects on
nicotinamide, NAD+ and methyl-group metabolism. Cell studies have
reported increased intracellular NAD+ and changes in SIRT1-related
signaling. These laboratory findings do not establish a health or
weight-loss effect in people.
Research has also examined NNMT inhibition in diet-induced obesity
and aged-mouse models. Results from those models cannot establish
human effectiveness, an appropriate human dose or long-term safety.

Preclinical Research
What Studies Have Examined
NAD+ and SIRT1 Pathways
In adipocyte models, 5-Amino-1MQ has been reported to increase
intracellular NAD+ levels by approximately 1.2–1.6 times and to
support SIRT1-related activity. These are cell-model findings, not
demonstrated effects in humans.
Adipose-Tissue Metabolism
Diet-induced obese mouse studies reported reduced fat mass and
body-weight gain without reduced food intake. Researchers examined
changes in adipocyte energy expenditure, fat oxidation and
white-adipose-tissue characteristics.
Glucose and Insulin Markers
Animal studies have reported changes in markers related to insulin
sensitivity and glucose tolerance. These findings have not been
confirmed in human clinical trials.
Aged-Mouse Muscle Studies
Research in aged mice has examined grip strength, running capacity,
muscle function and regeneration. These results remain preclinical
and do not establish effects on human strength or recovery.
Research Hypothesis
NAD+ Precursors
Because NNMT acts on nicotinamide metabolism, researchers have
proposed that NNMT inhibition could complement NAD+ precursors such
as NMN or NR by reducing the consumption of related building blocks.
This proposed combination has not been established as a clinical
benefit in people. No human trial has confirmed its effectiveness,
safety or interaction with NAD+ precursor supplements.
Evidence, Safety & Regulatory Status
Limitations and Research Status
Available evidence is preclinical. Cell and rodent findings have not
established whether 5-Amino-1MQ is effective or safe in humans, and
animal findings cannot determine a human dose or predict human
outcomes.
There is no formal human safety database. Animal studies have not
highlighted major acute toxicity at the doses studied, but long-term
effects, potential interactions and appropriate human use remain
unknown. Mild gastrointestinal effects or changes in energy have been
reported anecdotally; these reports are not a substitute for clinical
safety data.
5-Amino-1MQ is not FDA-approved for any indication and is sold as a
research chemical. Product identity, purity and quality may vary
between suppliers. Research status does not establish suitability for
human consumption or clinical use.
